Hello All,I'm currently researching the mitochondrial Aldehyde Dehydrogenase 2 (ALDH2) enzyme. ALDH2 is a non-P450 detoxifying enzyme that works to convert aldehydes to their corresponding acid. ALDH2 catalyzes the NAD+-dependent irreversible oxidation of a wide spectrum of endogenous and exogenous aldehydes.ALDH2 encodes a mitochondrial matrix protein that is constituitively expressed in a variety of tissues including liver, kidney, heart, lung and brain. ALDH2 is the primary enzyme involved in the oxidation of acetaldehyde (Km < 1 μM) during ethanol metabolism.My question for you all is what is the best way to probe ALDH2 in melanocytes? I am currently running Agilent Mito Stress tests on a few melanoma cell lines but I would love other ideas on how to flush out more information regarding ALDH2's role in the mitochondria of melanocytes.
