particle analysis using ImageJ

Hey, all, could you help me with the following problem? I will appreciate it very much. I tried to use the plugin "Particle Tracking" of ImageJ to analyze cell movement. Cells were not stained and Images were captured with light microscope using time-lapse recording. However, in the application of ImageJ for these images, it is difficult to detect cells. I tried to process the images so that the background became dark and cells became bright. In this way, cells could be detected by the software. However, when linking these particle movement to generate trojectories, these trojectories are not continuous. For example, the trojectory for one cell was interrupted at some place. How could I do to improve this analysis? Thanks.
