Epidemiological and clinical studies provide evidence that type 2 diabetes is a pro-inflammatory state with increased circulating inflammatory markers that track with hyperlipidemia. Data from animal and human models indicate that heart failure in diabetes or diabetic cardiomyopathy is associated with ectopic lipid, ceramide accumulation and inflammation in the heart. This project will use mouse models of diabetes and metabolic cardiomyopathy to test the hypothesis that ceramides are generated in the lipid-overloaded heart where they serve as modulators of tissue damage.
