 The role of the lipid environment in the pharmacological and biophysical properties of the D2 receptor

Lipids make up more than half of the brain in dry weight and a wide range of psychiatric disorders are accompanied by disturbance in lipid metabolism – n-3 polyunsaturated fatty acids (PUFAs) in particular. Yet the role of lipids on brain function has been overlooked. N-3 PUFAs are main components of cell membranes where they modulate biophysical membrane properties and consequently transmembrane receptors’ function. Recent data obtained by the host lab show that n-3 PUFA deficiency particularly impacts signaling downstream of the dopamine D2 receptor (a G-protein coupled receptor GPCR) and associated behaviors, which are altered in various psychiatric disorders. The goal of the project is to determine how membrane lipid composition, and PUFAs in particular, impacts the pharmacological and biophysical properties of the D2 receptor. Few studies in the literature have focused on this aspect due to the inherent difficulties of isolating and reconstituting GPCRs in functional form and the lack of experimental approaches with enough high sensitivity. For such, plasmon waveguide resonance (PWR), a technique recently installed in the laboratory will be applied that allows one to follow lipid reconstitution, ligand binding-dependent activation of the receptor and recruitment of downstream signaling (e.g. G-protein interaction) directly and with high sensitivity (femtomole quantities can be detected). The use of model lipid membranes of varied lipid composition, namely concerning n-3 PUFA, will shed light on the modulation of receptor signaling by lipids.
A candidate with background in Biochemistry/Biophysics is desired.
