I have a problem in doing bacterial agglutination titre of normal fish serum.
After 2 fold dilution in PBS in U shaped microtitre plate, I am adding 50 microlitres of formalin-killed Aeromonas hydrophila (OD- 2)...
This procedure was working previously, but now not working. Is there any problem in the plate...or bacteria.
Should I use sodium thiosulphate to wash the plate... or any other solutions please.

You said that your procedure worked previously. Have you changed anything recently before it stopped working i.e. changed a supplier of serum or culture plates.
Also, are you using a new stock of bacteria or one that has been through a large number of passages?
