i know that when protein kinase C is activated, translocates to the membrane where it is bound and phosphorylate surrounding molecules. But i wanted to do an activity assay and to get maximum level of activity i was thinking of doing an extraction step which will dislocate the enzyme and release it in the cytosol, which could be measured. SO my question is do u think that by extaction step, the pkc will be rendered inactive and i will be measuring non- active enzyme in the end??? Does that make sense? I am very new to this area so help me out please!!!
