what's the relationship bewteen CPS and RLU in Chemiluminescence

I have performed the SEAP Reporter Gene Assay by Chemiluminescence Detection Using Luminometer Plate. I get the 5 Second luminometry reading(CPS, count per second). I do not fully understant what the relationship between CPS and RLU(relative light unit) and how can I change the CPS to RLU.

Could you please provide the name of the company and the type of reader you are using as well as the manufacturer names and products? so I could understand the system you are working with.

Different readers give widely

Different readers give widely different readouts in terms of their (arbitrary) units. The Perkin Elmer V3 for instance gives readings in hundreds of thousands of units, whereas an older one (forgot make) used to give low numerical values in a four decimal place format. Irrespective of the way you get these values, the averaged value (in your case over 5 seconds) is used to obtain the RelativeLU by normalizing to the control condition values.

our lab bought a new

our lab bought a new luminometer from perkinelmer, I used the injector to add the reagent, but I found 1.5 ml reagent went to the waste bottle without doing assays, Is it normal?

Hi Ziseputi
When injector is used to add reagent first step will be priming. During which reagent will be piped through tubing till the end i.e plate. The total length of tubing varies depending on instrument make. ( in your case it is 1.5 ml) But this is not wastage. You can collect and reuse it provided collected it in clean plate and before priming tubing is empty. Mostly such plate will be provided with instrument.
