Is there any simple method to demonstrate demyelination in peripheral nerves using formalin fixed paraffin embedded sections other than H&E ? Toluidine blue staining seems to work only in OsO4 fixed resin embedded sections. Can anybody give me the protocol for Kulchitsky Pal myelin method ?

 Since you have embedded the tisue in to paraffin, the fat in the myeline is removed too. For peripheral nerve, if you don't think histological evaluation is enough, you may want to try an antibody against MAG (Myelin Associated Glycoprotein). It is involved in the peripheral demyelination process in several diseases. However, it is important to understand the underlying pathology to be able to detect demyelination and evaluate your tissue. Is it caused by inflammation, autoimmune, tissue damage?? All this palys arole in picking the right detection method.
