The response factor for any analytical instrument is simply the ratio of the response from the instrument to the concentration or mass that produced that response.

For chromatography,

RF = peak area/concentration injected (or you can use peak height, or mass injected)

Response factors are used to calibrate the response of the instrument.  If you know the RF for a compound, and you measure the response (area or height), you can then calculate the concentration (or mass) that was injected.
