Hi Does any one of members worked with expression of Monolconals in CHO .I could not able to express them to get some decent expression.I use singe and double gene vectors into modified pcDNA3.

Yes I am making enough antibody making cells.I use pcDNA3 for transfecting the CHO cells using electroporation.I have modified pcDNA3 to make them into single vectors and double gene vectors .single gene vectors contain heavy chain with the selection markerand heavy chain and light chain and double gene vectors contain light and heavy chain with the selection marker(DHFR)
We successfully expressed heavy and light chain genes by co-transfections of CHO cells and then purified the IgG. We offer this as a service. See www.genovac.com for more information

I've never tried to express mAbs in CHO cells but I (and colleagues) have had problems with pcDNA3.1.

I'm not sure I understand this. I guess you're doing stable transfections (electroporation) and selection etc to get enough Ab producing cells...?
