Hi,

I recieved an email from someone asking about a matrix for the Y-filer ABI kit. I'm assuming it was an email not a response to a topic I've subscribed too but I deleted the mail by mistake so I hope that you read this.

A matrix is used by capillary electrophoresis machines to discriminate between the different fluorescent dyes used in a particular assay, and to prevent spectral pull-up in your traces. Depending on which kit you're using, you need to tell the machine which dye set (matrix) to use.

In the case of the y-filer kit, the matrix you should use should be listed in the kit. Unfortunately I don't have access to a kit at the moment (I haven't personally used it) but I think it's dye set/matrix E or F. Your best bet is to call ABI tech support for your country.

You will need to purchase a matrix kit separately to the yfiler kit and run an injection of matrix on the machine to calibrate it for that dye set before running your samples. This will enable the machine to view the emissions data and apply this information to the spectra from your samples.

Some matrixs have different kits for the 3100 and the 3730 so you should ask ABI which one to purchase for your needs.
