There are 47 protocols I found for plasmid prep and cloning. http://www.e-biotek.com/index.php?option=com_content&view=article&id=165:plasmid-protocols-21-30&catid=35:dna-methods&Itemid=55I need some protocols for plasmid cloning selection, hope somebody can give me a favor.
Can you give us some more information on what you are trying to clone into which plasmid? Are you running on a tight budget or can you afford the commercial kits from companies like Qiagen, Biorad and Promega? For plasmid purification the gold standard against which most kits are compared are usually the Qiagen kits. But you may have a personal preference according to scale, cost and the quality of the prep at the end of the protocol. Also look through our protocols pages at http://www.scientistsolutions.com/Science-Protocols.aspx and see if anyone has posted something useful for you
Thank you for your reply. We don't have problem with budget, but the molecular cloning is just at the beginning.I try to clone PPAR-gamma into a vector with GFP which I purchased from Clontech. I will further transfect the PPAR-gamma cloned vector to some mesenchymal stem cells that were established at our lab.I used to use products from Invitrogen for our molecular biology experiments. I will let you know which kits I will buy after reviewing your products. Again, Thanks.
Are you working in Signapore?
Origene already have various plasmids with PPAR gamma and GFP. If time is more valuable than money:http://www.origene.com/cdna/search-all.mspx?term=ppar+gamma&product=ORFCLONE&go.x=0&go.y=0
