Hi
our lab always use cleanup kit for DNA and vector purification from agarose gel. Some other lab use DNA and vector directly from gel without purification. Can anyone tell me the protocol for this kind of ligation?

Does this method use some kind of agarase?

We also use a clean up kit - Gene Clean Turbo is so fast it's hard to believe that there is a faster alternative
