Hi Amrtiha,

Here are some websites (and a video) on the basics of gene cloning.
The sites offer links to more specific instruction.

http://biotech.about.com/od/cloning/tp/DNAcloning.htm
http://ghs.gresham.k12.or.us/science/ps/sci/ibbio/biotech/genecloning.htm
http://users.rcn.com/jkimball.ma.ultranet/BiologyPages/R/RecombinantDNA.html

and a video
http://highered.mcgraw-hill.com/olcweb/cgi/pluginpop.cgi?it=swf::535::535::/sites/dl/free/0072437316/120078/micro10.swf::Steps%20in%20Cloning%20a%20Gene

Additionally, there is a subcloning protocol under the molecular biology heading in the protocols section of the website (found just under the SSI banner). That should also be helpful.
