Hi.. I have been doing PCR's for past three years but since I have switched to Roboscreen quantification kit, I am having trouble with my extraction Neg Conts. No matter how carefull I am, I'm getting copy numbers in my NCs, bcuz of this I am unable to justify my low copy number samples. My senior keeps saying that it is my handling problem.... Without ne further explaination I have seen the same prob in her PCR also. Please if ne one can help me to get rid of this problem.

do it in the reversal manner. take different kit.

Well I have done that also and the problem goes away.. If I put my ENC in another rack, far far away from the samples; in other word cheat, ENC comes undetected. I tired HCV and HBV Quantification kit and the ENC's r never clear normaly, only when I change the Threshold. For CMV and EBV quantification the ENC's are always undetectable...
