Hello ,
to everybody iam working under the DBT project ,i need to culture mycobacteria strains which are slow growers. and i had a doubt whether i have to use liquid media or solid media. can any one suggest the better one with faster growth.

What are the sources of your mycobacteria cultures? Are the stock cultures at 10^9 cfu/mL?

If so, then streak plating these on Middlebrook 7H10 or 7H11 agar w/ 10% Middlebrook OADC supplement should demonstrate growth within 7-14 days depending on the strains.

If the source of your cultures are low level environmental strains, then I would suggest incubation in Middlebrook 7H9 broth w/ 10% ADC supplement. Incubate ~ 100 ml broth cultures with continual shaking (~200-250 rpm) in 250 mL flasks.

Remember that all Middlebrook supplements (OADC or ADC) should be added after autoclaving (at less than 50 C).

Another consideration should be what are your intention with your cultures after they have been grown up. It may be easier to use one or the other depending on your applications.
