Does anybody have an experience with immonoflourecent CD14 Ab on human frozen sections?

What tissue/treatment is it? There is quite a lot of auto-fluorescence - the methanol quenching step is critical. Additional post-fixation also helps.

I didn't use methanol but I can try.
which clone/company of CD14 did you use?

I think I used the Biolegend biotinylated Ab with Strep AF633 (you'll need the appropriate laser (HeNe?)/filter cube combination. In general, using the red dyes reduces auto-fluorescence with many human /mouse tissues.
Are your sections acetone fixed/flash frozen?

I fix tissue in formalin before freezing. Usually I don't have problem with most of fluorescent Abs but I am not sure in my results with CD14.
