Has anybody used the Pierce (Now Thermo scientific) Sulfolink Ab immobilization kits? Do those work better for small peptides (e.g. ones used for Ab affinity purification) than the carboxyl-crosslinking systems? And finally, does the cysteine based background quenching affect protein binding properties (again, for e.g Ab-peptide interactions)?
Just for the record, the SulfoLink kit worked pretty well, with nearly 65% of the loaded peptide being immobilized. Between 50-80% of non-specific binding (depending on cell type: the original Ab showed no difference, now I see distinct expression differences between neutrophils and monocytes) was eliminated, based on FC assays with different dilutions/blocking peptide.
