We are having problems finding/using an extraction protocol of maize roots that consistantly gives us usuable DNA mitotic cell cycle peaks. We have tried numerous buffers, and either our cell count is to low (350 cells in 3 minutes) or there is only debris. Does anyone have a protocol that works well on this tissue type?
