Hello!
I want to know is there any kit available for elution of DNA bands from Polyacrylamide gels???
I have used Qaigen and Sigma gel extraction kit but i am not getting good recovery, consequently my down stream processing is hindred.
Any new protocol for elution of DNA bands from Polyacrylamide gels is also welcome!!
Looking forward to hear from you all at yr earliest.

I used to elute protein from PAGE gels and one of the methods I used included a first step in which the gel slice was cut up into small pieces and put in to an eppenforf tube with 200-300L of buffer. The protein moved out of the gel, into the buffer.

As you know when using agarose gels, DNA (especially short molecules) tend to difuse out if you switch off the power supply. Maybe you could elute it in a similar way - into water or TE. I guess it would be more efficient if you used a PAGE gel that was low percentage with little cross-linking.
