Has anyone successfully performed staining on paraffin embedded sections using ChAt antibody (Chemicon AB1582) with fluorescent secondary? Did you use any antigen retrieval methods? Our first trial (no antigen retrieval) with rabbit and rat nerves showed low level, non-specific staining.

I've used Chemicon AB144P on frozen brain sections. The background looked a lot better when I opted out of antigen retrieval (Vector Labs) and just incubated the primary for 48hrs at 4C.
