Hello, Does anyone have the experience on this tube formation assay in 96-well plate? I just started to try HUV-EC-C cells on the assay by Chemicon Kit (Cat# ECM625). It worked well at first time. But, when repeating later, it did't work. I don't know what happened even I did it exactly the same way. Is it mostly possible the cells? Thanks.

It might the BME itself. Matrigel, Cultrex, etc. are notorious for their lack of consistency from batch to batch. There are numerous companies out there with different angiogenesis assays that you might want to try out.

Glycosan BioSystems recently contacted me in regards to beta testing their anti-angiogenesis product. I didn't take them up on it, but they said their stuff is defined (so you wouldn't have consistenct issues). It is a beta product, but it's also free.

Glycosan BioSystems recently contacted me in regards to beta testing their anti-angiogenesis product. I didn't take them up on it, but they said their stuff is defined (so you wouldn't have consistenct issues). It is a beta product, but it's also free.

If you have not come across a suitable assay yet, Vivo Biosciences may be able to help. Our assay is reconsituted in HuBiogel, which does not contain any detectable growth factors. This offers an element of control not available with Matrigel since it contains GFs.

HI Seagull,
I was wondering whether or not you stuck with the Chemicon angiogenesis kit? Because I ordered it then I came across your post ... read the replies and thought 'oh no!' and I've not done tube formation assay before! So could you kindly let me know what happened in the end? Did you purchase something else and did it work?

Hi Jabba and VBI, thank you very much for your recommendation. I should try them, but haven't yet.
Yes, bubbleT, Chemicon angiogenesis kit is the only one I have used. I found that the results were quite unstable, sometime good, sometime not. I have discussed with several experienced people. Accoeding to their experience, besides the matrigel, the condition of the cells is also one of the important fact that affects the result. It really is. In my experiment, the early passage cell usually had better tube formation. I am thinking to try Glycosan BioSystems that Jabba recommended.

Once you have ordered and tried the Glycosan BioSystems a couple of times, can you let me know how you get on? As I am interested in knowing because if results are reproducible then I will consider trying/buying this product also.
