Does anyone have a good protocol for concentrating recombinant VSV-G pseudotyped lenti particles?

There are two ways. One is ultracentrifugation which results in loss of titers but allows you to concentrate your virus by factor of 100. On the other hand, there are the Amicon Ultra-15 columns by Milipore that I have used andthey work great. They concentrate your virus prep by at least 30-fold.

How long do you spin the sup containing virus?

I have used both of these methods before but found the recovery of virus from the filtration device quite poor and resuspension of virus post-ultra-centrifugation to be difficult and to result in loss of infectivity.

Do you use sucrose in the ultracentrifugation and if so at which concentration and volumes etc?
