Hi

I found this article using STZ in distilled water in the induction of type 2 DM. I did the same procedure and it didn't work out so if there is anyone working in this felid please send me the reference that you used in your successful Type 2 induction

I'll be thankful if you did.

You wont induce Type 2 diabetes with STZ. STZ kills beta islet cells, which results in Type 1 diabetes. There are several ways to utilize STZ. The most widely accepted protocol is i.p. injection of 50 mg/kg STZ in NaCitrate pH 4.5 made fresh each week (pH with 1M Citric acid). You must inject the animal on 5 consecutive days. Animals will begin to show high glucose levels after 4 weeks.

Hi,

Rb has cleared most of the things;

The pathogenesis of type 2 diabetes is complex involving progressive development of insulin resistance in liver and peripheral tissues accompanied by a defective insulin secretion from pancreatic beta cells leading to overt hyperglycemia (an abnormally high amount of glucose levels in blood)

With the help of this, you can add some more options such as neonatal diabetes, to produce this you have to inject low dose of STZ (~35 mg/Kg, can very based on lab to lab response) to the pups around 4 days or later but not after 21 days. It will lead to the beta cell killing which will cause mild diabetes to the animals. And due to long time exposure (exp age will be more then 10 weeks) of diabetes to the animals insulin resistance will also develop.

Let me know if required further information

Chintan
