I have a very good transcription efficiency with CMV promoter. Currently I am using pFLAG-CMV-2 and it is working just great.

Has pFLAG-CMV-2 got only CMV promoter on it? Or any other promoter?

Most people got great results with CMV promoter, but there is few articles about EF-1 α promoter. So, I want to ask this question in this forum.

I am glad that the answer that you got helped you in your research.
I will read more about pBudCE4.1. Fortunatly the vector that I am using is very good for my protein expression.

Thanks for these wonderful words. I like sharing my ideas with other scientists, because we are all trying for the same thing I suppose: To get the best results in our experimental designs. A suggestion may help a lot, I believe in it. (By the way, sorry for my English. I hope you can understand me easily.)
And my research: I'm trying to clone two HIV proteins, they are about 400 bp, very small as you can realize, but they have important roles in HIV regulation. I choosed vector pBudCE4.1, because it is a mammalian expression vector, and I want to see the effects of these proteins on human cells. I cloned one of them yesterday, I hope I can get good results like you.

Yes I looked at the map. But I prefer the one that I am using now that have already an N-terminus Flag tag.
