What do you use for making "wells" around tissue sections in order to minimize the amount of antibody needed to stain a section?

Use a pap pen. It may be called Liquid Blocker or Liquid-repellent slide marker pen for staining procedures.

If you are in the US, you can order a 'PAP pen' from Fisher or VWR or Sigma - most bio-medical suppliers should carry it. Just shake it, and 'draw' the 'wells' you desire!
