Hello,

I'm trying to work out a protocol for ChIP with TAP tagged proteins.
Now, I'm confused about NaCl concentration in cell lysis buffer (which is later used for IP with IgG bids).
In several protocols that I read, the NaCl con. goes etween 150mM to 500mM.
My supervisor is not sure what to concentration to use.
Can anyone help me?

150mM NaCl is considered the default "low-salt" IP buffer in most ChIP protocols. A high salt wash is often used @ 500 mM or higher, but make sure you do not exceed the salt titer of the antibody you are using otherwise you will loose all your signal.
