Dear all,

I am trying to perform T cell proliferation assays using macrophages as APCs. It does not seem to work, becuase I used live macrophages. So I decided to use killed APCs as accessory cells. I used mitomycin and it does not seem to work too. Is irradiation the best solution. I am using CFSE stained T cell responders.
Can anyone working with macrophages suggest the best protocol for proliferation assays? Thanks
