Could someone tell me if it is worth quantifying antigens in paraffin sections using immunofluorescence? How many sections are needed, say with a heterogeneous antibody distribution such as in skin, and how do I safeguard that all unmasking the epitope in every sections would be uniform?If large numer of sections are needed, how to safeguard against quenching of fluorescence? sknman
