i am failing to get full sequence of my pcr products even when i ask them to purify before running the sequencing reaction. i lyophilise the pcr rection mix and send it. i tried cloning my amplicon to a T/A vector but didnt succeed. can anyone suggest me a good sequencing service which is both efficient and economical. my amplicon size is 1.6kb. thanks

Shinysangma: I have currently been looking for sequencing contract labs as well. The two that I have found to be both economical and efficient are: Molecular Cloning Laboratories - South San Francisco, CA Eton Biosciences Inc San Diego, CA If this post has helped you solve your problem please let Scientist Solutions know about it by posting a success story at http://www.scientistsolutions.com/t1262-Success+Stories.html or email marcia@scientistsolutions.com. By 'full sequence' how much sequence are you getting? Are your sequencing primers the same as your PCR primers or nested? You need to keep in mind that sanger sequencing reads don't often give useful base calls after 7-800nt. So in a best case scenario, assuming a few reads from each strand, you 'might' get overlap between your 5' and 3' reads. Ryan
