I want to know what would you regard as a change in intesity between two gels of similar group,. Is 2 fold change is significant.

First you'd have an internal standard otherwise any quantitative comparison is pointless between gels. Then, the "change" in intensity is up to you. Some software has a default 1.5 fold.
