Two early applications of subtractive technology for comparison of transcript profiles were the identification of the genes that were activated at the gastrulation stage of Xenopus laevis (1) and cloning of T-cell receptor (2). In the following years a number of applications and modifications of the technique has been described (3-5). The incorporation of polymerase chain reaction (PCR) into the procedure caused dramatic increase in sensitivity and made it possible to use for subtraction as little material as it's available from a few cells (6-11).
