Hi All,
I am using Lipofectamine for siRNA transfection of Hela cells.
I have got some problems with cell death, I guess the toxicity of the transfection reagent. ANy one had problems like that before?

I have had seen similar issues with lipid reagents. I use GeneJammer from Stratagene - it works well and it is a lot less toxic.

I was transfecting siRNA duplexes into some glioma cells and encountered difficulty. After trying several transfection reagents, I found Lipofectamine RNAiMAX to be the best for my cells.

For a quick check of transfection efficiency and cytotoxicity Ambion has a kit for easily assessing GAPDH knockdown. I found it very useful for optimizing my transfection protocol.
