Hello everyone,

I am currently working with human primary cells that are really to come by. I am hoping to do titration of the antibodies I am going to use for FACS analysis. Because the primary cells are really hard to get and I don't want to waste them in titration, I am wondering whether it is possible to titrate the antibody using a cell line instead of the cells that I will actually work with in the end?

Any suggestion would be great. Thanks!!

It is easy to work with cell lines obviously at the beginning. But you have to know the difference in the gene expression between your primary cells and the cell line. Check the literature to see if some groups already reported the expression level of your gene in these cells.

If your cell line expresses the antigen then there should be no problem.
