I need advice. I have to clone 1645bp fragmeni into expression vector. I tied to clone my gene into pQE32 and pQE 70 vectors from Qiagen, but there was no overexpression. I've decided to change the vector and now I wonder which one I should use: Champion pET Directional Topo Expression Kit or pBADDirectional Topo Expression Kit. Has enyone some experience with those kits?

Have tried changing the strain of the bacteria you are using to improve expression?

E. coli M15 works well with pQE32

Did you feed your cells enough IPTG to promote the expression as your gene will be inder the control of a lac repressor?
