What is the detection limit for protein identification on 1D gel bands or 2D gel spot samples?



The detection sensitivity of MALDI-TOF/TOF and 4000 Q Trap (LC/MS/MS) is around 10-20 fmol for standard protein digest in solution. For 2D gel spots, if you can see it with coomassie blue, it is highly likely we will be able to identify it. If you can see it with Sypro, there is a chance, but the low stain spots will not be possible. For 1D gel bands, however, it will depend on the protein complexity on the gel band. For SDS-gel samples, if a band containing a single protein is visible with colloidal coomassie blue stain, there is probably enough material present for protein ID.
