Do you know of a very critical effect of pH on binding of immunoglobulins from different species to ProteinA-Sepharose beads in immunoprecipitation? If so, could anyone suggest the optimal pH for mouse IgGs? Thanx

Check out this link for info on which antibodies bind best to which protein (A, G, L etc.) and what pH is best. It looks like pH 7-9 is best for Protein A.

http://www.piercenet.com/files/1601323%20Antibodies_2.pdf

and this link shows what pH is best for the different proteins

I have had new, they tell:
option pH for protein A or G is 7-9. but with to immunoglobulin, you should be balance pH of the fractions after elute. :D

piercenet is instructive!

Please note that mouse IgG1 will bind to protein-A weakly. (Trust them!)

Try something else.
