Dear Forum members,
I am working on the erythromycin producing by Saccharopolyspora erythraea which is from USA, now have some questions. Production media is soybean powder 2.0%,yeast powder 0.5%,(NH4)2SO4 0.032%, bean oil 6.0%,CaCO3 0.75%, CuCl22H2O 0.0035%. The mycelium will autolyze after 120 hour during fermentation.

Can you describe your question in more detail? Are you concerned with controlling the autolyze characteristics of your fermentation culture?

Yes, The autolyze is not what I want.
The natural cycle of fermentiation should be 180 hour,but it often autolyze in 120 hour, and the yield is only 6.0-6.5g/L.
I want to increase erythromycin productibity to 8.5-9.0g/L.But I don't know what is wrong with it.

Any help would be appreciated

I'm not sure what organism you are using but the following review article gives quite a few suggestions about maintaining a stable transformant for biotech fermentation manufacturing.

Schallmey M, Singh A, Ward OP. Developments in the use of Bacillus species for industrial production. Can J Microbiol 2004, 50: 1-17.
