2D gels too brittle [View Printable]

Hello folks, We make our own SDS PAge gels for 2d. Recently I started filtering Acrylamide and bis solutions and i notice that the resolution has improved but that the gels are very brittle . Wonder if there is a work around or something. Thanks

Hi, I did alot of 2D gels by my self. Tell me what is the problem what do you mean by brittle?

Hello , We separate the top glass and gel is on a silanized glass plate and we scan these images. I noticed however that after fixing the gels 2 out of 4 gels broke to pieces. I have never ever seen this before. The glass plate is fine . Subha

Once I had a problem with gels that broke. What kind of fixing solution you use? What are the staining that you are using?

Hello gsovac. I use 30% ethanol and 10% acetic acid as a fixing solution. I fix for 5 hours and change every 4 hours. I know this is probably too vigorous. This has given me great images though. We use Cy dyes / fluorescent dyes for our samples. Thanks for help

I have used 30% methanol and 10% Acetic Acid. But also instead of 10% AA I have used 7%. Try to change the ethanol to methanol and 7% AA and less changes of the fixing solution. Guy
