Hi we are working with trying to identify changes in the phosphorylation status of proteins with after we knockout our novel protein. We wanted to do it via mass spec after we performed a 2D gel and then transfered to a membrane. Then we wanted to probe with either anti-phospho serine and then also anti-phospho threonine antibodies. Does anyone have a protocol for idenitifying proteins from a membrane via mass spec? Or could they refer me to a website or something that could help us out. Thanks. :)

Hi, do you mean you want to use the membrane from a blot to extract the protein after identification with antibody?
