Hello,Has anyone worked with transplatin in cell culture tox assays and found it to be toxic ?I'm working with the drug (300 micromolar) using short incubation times (2 hours) in ovarian cancer cells. I'm getting reproducible IC50 values 24 h later of around 150 micromolar. The drug is dissolved in EBSS by vortexing and brief sonication using a probe sonicator on low setting. Many thanks for any thoughts
Though I dont know your area very much. 300uM of any drug treatment looks to me quite a high in concentration
Yeah, it's a short contact time and it's standard practice for this FDA/EMEA approved protocol that I'm using (Top conc = 100mg/ml, which is about 300 micromolar drug).
Hi Delilah, check out this paper:http://www.ncbi.nlm.nih.gov/entrez/query.fcgi?cmd=Retrieve&db=PubMed&list_uids=8396390&dopt=Citation
Thanks for that info.
