2 Dimensional Thin Layer Cellulose Electrophoresis of Phosphoamino Acids

Overview

This protocol describes the electrophoretic separation of phosphorylated amino acids on Thin Layer Cellulose. This procedure often follows metabolic labeling of cellular proteins by 35S or 32P and tryptic peptide digestion of polypeptides (see Protocol ID#310). This procedure requires a horizontal flatbed platform, with electrodes embedded in reservoirs at the ends of the platform, and cooling tubes for running cold water underneath the flatbed. The phosphorylated amino acids can also be analyzed by High Pressure Liquid Chromatography (see Protocol ID#591).
