Hi!

I'm looking for an antibody directed against a protein resident in golgi vesicles. Any suggestions? I want to use it in immuno fluorescence and western blot.

Thanks

Hi
People in my lab used TGN46 for immunostaining, and it works perfectly.
It isnt good for WB.

You should try and obtain a Y/CFP-tagged version of the N-linked glycoprotein-processing enzyme galactosyltransferase (GalT)

example of its use:

Golgi inheritance in mammalian cells is mediated through endoplasmic reticulum export activities.
Mol Biol Cell. 2006 Feb;17(2):990-1005.
http://www.molbiolcell.org/cgi/content/full/17/2/990

actual cloning paper:

Sciaky, N., Presley, J., Smith, C., Zaal, K. J., Cole, N., Moreira, J. E., Terasaki, M., Siggia, E., and Lippincott-Schwartz, J. ((1997). ). Golgi tubule traffic and the effects of brefeldin A visualized in living cells. J. Cell Biol. 139, , 1137-1155

If you still need to do immunotechniques you can use the anti-GFP antibodies that are commercially available.

Any paper from the Lippincott-Schwartz lab should help you out because she is so preminent in this field.

Hi Jenny, I would contact someone at the glycobiology program at the university of california in san diego. Since it is a golgi protein (and the golgi is heavily involved in glycosylation) i thought that someone there might be able to help...

good luck!

k.
