Can we measure the real time binding using a surface plasmon resonance state of the art technology? A little description of the protocol will be helpfull.

It's been awhile since I've thought about or used SPR, but from what I remember, analyte and ligand association and dissociation constants can be determined and therefore the rate and equilibrium constants.

Yes, SPR does measure real time binding - depends on what experiment you are trying to set up, though, to be able to say whether it will work for you. Basically one component is immobilized on a chip, and the other is flowed over it. When there is binding, you get a signal.

Simple enough, right?

Exactly. SPR is simply a refractive index detector (mass) that gives results in real time. Unlike ELISAs, you can see HOW fast your relative on and off rates are instead of just knowing the net.
