Hi every body Can any body help me for my protien modfiacation, I myc tag my protein and did westran , I saw three bands of my protein , two very close and one 15 kd down. This happen to my protein when my yeast cell undergo meiosis. its strats from time point 4 in yeast meiotic time course. i don,t know what type of modifacation is it? can any body help? Cheers Mk

You could try treating your cultures with N-glycosydase F to look at glycosylation This drug cleaves asapargine linked glycan chains on glycoproteins. I agree with Frasermoss. Yeast are very good at putting glycosylation on proteins. Check your protein sequence for Asn linkage sites for N-glycosylation (Asn-Xaa-Ser/Thr) before you buy the digest kit though.
