Hello, sorry in addition to all that useful information, does anyone know the largest area you can photo bleach, and if you are using a confocal system, does it photobleach on one specific plane? or am I able to bleach a whole area of a cell?

Depending on your system, you are able to select the area that you want to bleach by drawing on your image with an area marker in the computer imaging program. The area that you bleach is determined by your image size on the screen. It is generally accepted that you will bleach a portion of the area on the screen to analyze the bleached area and the area that is not bleached. Recovery of the bleached area depends on dye transfer from adjacent cells--therefore you want to have adjacent cells that are not bleached
