 HI all..

I did western today and notice that I saw only  5  (from 6 ) - 50 kd  bands of the marker.

in addition, in the ponceau I saw bands only untill the 50 kd marker.

meaning, I dont see protiens that are smaller than 50kd.

what could be the problem?

There can be multiple reasons or it:
1) you are using a membrane wit high pore size and transferred it for long duration leading to escape of your LMW proteins

2) A bubble or air space between your gel and membrane

3) Ponceau has alot lower sensitivity in comparison to CBB g250 and as a result youre unable to detect proteins at this range. Increase the loading amount and you may be able to visualize them.

4) degraded proteins in LMW region
