Did anyone do western blot of cytosolic / mitochondrial cytochrome C release?

I transferred the protein in semi-dry blotter for 40 minutes and the band was very weak (from ECL detection and exposed for 10 minutes).

Could anyone give me some advice? Thank you first!

the expression of cytochrome C in Hela cell is enough for WB. May be the concentration of primary or sencondary antibody should be improved.
