Hello , im new here to this forum....i want to do 2D electrophoresis on my GST purified protein which has 2 basic degradation products. My question is :can I do it with GST bound to it? i mean wont GST effect the charge of the protein? thank you very much.

Dear Koyo,
There is a 2D forum under proteomics, just to let you know.
As for you question,
I did alot of 2D but never with an attached GST to it.
I guess that it will change the pI of your protein and also the MW.
How is it attached to your target protein?

thank you for your reply, i had inserted my protein into the GST vector to express it in E-coli. and now my recombinant protein has GST in it. I can cleave ot of course but im wondering if I can run 2D on it as it is.

Yes Koyo,
You can run the GST fused Protein on a 2D gel.
I am nearly 100% sure that the pI would be effected.
just run the protein without GST fused to it.
