A method for the real-time quantification of microRNAs (miRNAs) employing universal reverse transcription (RT).




Description

A new method has been developed which allows the real-time quantification of miRNAs and their predicted target transcripts. The method utilizes a universal RT-primer followed by reverse transcription, and then a transcript-specific strategy in the PCR. Parallel amplification of a miRNA and internal reference or target transcript is possible. The method is straightforward, needs no RNA size fractionation and involves routine enzymatic manipulations.

Confirmation of computer predictions of 100s of miRNAs.

Quantitative analysis of the expression of miRNAs and their predicted mRNA targets.

Main Advantages of Invention

Universal reverse transcription of miRNA and mRNA allows for examination of both miRNAs and predicted target(s) from the same sample.

Fully operational for a number of cancer related miRNAs and target mRNAs.
